VKORC1
Vitamin K Epoxide Reductase Complex Subunit 1
What it does
The drug target for warfarin. Variants determine warfarin sensitivity.
Why this matters for Black patients
African-ancestry populations have lower frequencies of the VKORC1 high-sensitivity allele than Europeans, which means standard warfarin dosing is often subtherapeutic. CPIC has a separate dosing algorithm for African ancestry.
Common variants
| Variant | Phenotype | African | European |
|---|---|---|---|
| rs9923231 | Sensitivity-determining promoter variant | 0.100 | 0.400 |
